Preparation, characterization and antimalarial activity of dihydroartemisinin / ?-cyclodextrin spray-dried powder

In this study a factorial design was used to optimize process conditions produce spray-dried powder of dihydroartemisinin (DHA) with ?-cyclodextrin (?-CD). Predicted for maximum yield and minimum residual moisture were as follows: inlet temperature; 120 °C, aspirator setting; 100%, feed rate; 25%, molar ratio DHA:?-CD; 1:2. The higher percent inclusion obtained in the lower DHA:?-CD. DHA existed amorphous state 1:2 M DHA:?-CD whereas it crystalline 2:1 complex formation between ?-CD revealed by FTIR study. showed dramatically enhanced dissolution rate unchanged antimalarial activity. Moreover, chemically physically stable at 30 75%RH 3 months after storage.

Antimalarial activity of thiophenyl- and benzenesulfonyl-dihydroartemisinin.

Each diastereomer of 10-thiophenyl- and 10-benzenesulfonyl-dihydroartemisinin was synthesized from artemisinin in three steps, and screened against chloroquine-resistance and chloroquine-sensitive Plasmodium falciparum. Three of the four tested compounds were found to be effective. Especially, 10 beta-benzenesulfonyl-dihydroartemisinin showed stronger antimalarial activity than artemisinin.
